What they seem to conclude is that any mu agonist increases the cardiovascular effects of cocaine. I imagine that since the cocaine is potentiated, people who die from a speedball probably injected what is normally a good dose by itself and it turned out to be a cocaine od. If you read the whole article, they suggest a possible interaction between the opiod peptide system and dopamine (DA) levels in the nucleus accumbens (NA). Apparently, cocaine by itself has a positive effect on the opiod peptide system and heroin has an effect on DA levels in the NA.
"In the present study, we examined the effect of an acute administration of the selective suicide inhibitor of gamma-aminobutyric acid (GABA)-transaminase, gamma-vinyl GABA on increases in nucleus accumbens dopamine produced by a cocaine/heroin challenge in freely moving animals. Cocaine (20 mg/kg, i.p.) produced an elevation in extracellular nucleus accumbens dopamine of approximately 380% above baseline, while heroin produced only a moderate increase of 70%. Coadministration of these two drugs, however, produced a synergistic elevation in nucleus accumbens dopamine of 1000%. This response was reduced by 50% in animals pretreated with gamma-vinyl GABA (300 mg/kg, i.p.) 2.5 h prior to challenge. It is probably this huge increase in dopamine levels that increases the morbidity associated with speedballs. I would think that the amount of each drug used is the most important factor. Obviously, a much smaller amount of each will do. It's true that the two drugs blunt some of each other's effects, but mainly its just for the subjective effects (less anxiety, nervousness, shitty comedown from the coke, more awake than usual from the heroin).

Värt att ta med sig från denna text om man inte orkar läsa allt:
"any mu agonist increases the cardiovascular effects of cocaine". En "mu agonist" är alltså heroin, metadon, buprenorfin osv. och dessa potentierar effekterna som uppåt-preparatet (kokain i det här fallet)har på hjärtat.
I små mängder är det nog ingen större fara om man inte redan har problem men en av bieffekterna är alltså att det är mycket mer ansträngande för hjärtat och kan leda till stopp eller att man råkar överdosera lättare än om man tagit varje preparat för sig. Som någon annan påpekade. Bland annat så är det många kändisar som dött av det, en av de mer kända är River Phoenix.
Har själv överdoserat på detta sätt, hade tur som klarade mig men det var väldigt nära gravstenen.
Och innan någon påpekar, så är det kola och heroin som är en klassisk speedball, men principen går att applicera även på amf + heroin.

Det där är nonsens min vän. Det beror dock på vilken nivå man mixar, skjuter du heroin tillsammans med amfetamin eller kokain, ja, då är det farligt som fan, men det kommer inte ge serotonergt syndrom utan det kan försätta hjärtat i extrem stress och leda till en infarkt eller liknande - så det är det man behöver oroa sig för i det fallet. Men är man på nivån att man mixar ett par Conserta med valfri opioid så är man man rätt safe. Många, mig inkluderad, har opioider och ADHD-medeciner på recept och tar det dagligen, men det är så klart viss skillnad att vara ingången på något och att ta det utan någon som helst tolerans. Som vanligt så ska man inte ta för stora doser av något, tar du lite mindre så kanske du inte sitter och somnar.
Fast det brukar vara i slutfasen av ruset som man blir extremt trött och noddig, men som sagt aktivera dig och sitt eller ligg inte ned hela tiden.
Nej, det är absolut inte nonsens, läs på lite innan du svarar! Och jag förstår inte vad det är med folk, varför ska man hålla på att rekommendera sätt att blanda droger som är eller kan vara absolut livsfarliga? De som varit med ett tag och har koll vet att det är riskabelt att blanda droger.
Nu sa jag inte serotonergt syndrom men vissa har hakat upp sig på det - OK det är inte det man riskerar i detta fall utan s-syndrom är det man åker på om man överdoserar vissa droger som frigör serotonin t.ex. MDMA eller i vissa fall om man blandar vissa preparat, antidepp och MDMA t.ex. När man blandar olika preparat som frigör serotonin så är risken mycket större än om man bara tar ett preparat för sig självt. Det förklaras bra under rubriken "Neurotoxicitet" här:
